Cloning and Bioinformatics Analysis of the Gene Encoding Transcription Factor MYB44 of Sunflower (Helianthus annuus L.) under Salt Stress Conditions

Authors

Abstract:

Sunflower oilseeds (Helianthus annuus L.) are widely used around the world. Soil salinity negatively affects many morphological and physiological traits of sunflowers. Oil seed sunflower line tolerant to salinity stress (AS5305) was planted in normal and salinity stress conditions in a completely randomized design with two biological replications in a controlled environment. Salinity was applied in 8-leaf stage from NaCl source. About 24 hours after salinity stress, leaves were sampled. The MYB44 gene, as one of the genes involved in salinity stress resistance, was cloned in expressive vectors P35s-N and P35s-G, and bioinformatics analysis was performed on it. Analysis of cloned MYB44 sequence revealed more than 95% and 98% DNA and protein similarity respectively, between cloned gene with the MYB44 sequence in the NCBI, indicating high sequence conservation of this gene among different sunflower genotypes. The results of the present study can be useful in molecular breeding programs for the production of cultivars resistant to salinity stress and reduce the damage caused by this stress.

Molecular and Physiological Responses of Sunflower (helianthus Annuus L.) to Pgpr and Sa under Salt Stress

This paper presents the efficacy of PGPR (Azospirillum and Pseudomonas) and its modulation by salicylic acid. Two hybrids of sunflower (Hysun and Parsun) were inoculated with Azospirillum spp. and Pseudomonas spp. prior to sowing. Salt stress (20 dSm) was applied 28 d after sowing followed by foliar spray of salicylic acid (100 μM) after 4 h of salt treatment. Azospirillum and Pseudomonas inocu...

Cloning and characterization of a cDNA encoding type 1 diacylglycerol acyltransferase from sunflower (Helianthus annuus L.).

A full-length cDNA encoding a putative diacylglycerol acyltransferase (DGAT; EC 2.3.1.20) was obtained from sunflower (Helianthus annuus L.) seeds. The 1524-bp open reading frame of this cDNA, designated as HaDGAT1, encodes a protein of 507 amino acids with a molecular mass of 58.5 kDa showing high homology to DGAT1 enzymes of other plants. The protein characters, such as a predicted structure ...

Genetic analysis of proline concentration under osmotic stress in sunflower (Helianthus annuus L.)

Proline concentration has been often suggested as an indicator of osmotic stress. A better understanding of the genetics of this trait is however needed. In the present study, proline concentration has been assessed, together with root and stem growth, potassium, calcium and total soluble sugars concentration and stress injury symptoms, in seedlings of sunflower hybrids and their parents grown ...

Genetic analysis of leaf hydraulics in sunflower (Helianthus annuus L.) under drought stress

Improvement in leaf hydraulics is directly related to the improvement of plant tolerance to drought stress. Therefore, a field and pot experiment was carried out to determine the type of genetic variability and selection of parental types on the basis of combining ability for leaf hydraulics. Genotypes showed similar performance in both experiments; higher values were shown by drought tolerant ...

Improving Growth and Yield of Sunflower (helianthus Annuus L.) by Foliar Application of Potassium Hydroxide (koh) under Salt Stress

An experiment was conducted to assess whether KOH applied as a foliar spray could mitigate the adverse effects of salt stress on plant growth and yield of sunflower plants. For this purpose, KOH @ 0, 0.5, 1.0, 1.5 and 2.0% was applied as a foliar spray to salt stressed and non-stressed sunflower plants. Foliar spray of KOH increased the growth and yield under non-stress and salt stressed condit...

Genetic analysis and QTL mapping of agro-morphological traits in sunflower (Helianthus annuus L.) under two contrasting water treatment conditions

The present study was undertaken to investigate the genetic basis and map quantitative trait loci (QTLs), controlling agronomic traits in sunflower under well-watered and water-stressed conditions. Recombinant inbred lines (RILs) coming from the cross between sunflower parental lines PAC2 and RHA266 were evaluated in a rectangular 89 lattice design with two replications in each treatment condi...
